mRNA export: Travelling with DEAD box proteins
نویسندگان
چکیده
Recent studies have shown that the putative RNA helicase protein UAP56 and its yeast homologue Sub2p are not only involved in pre-mRNA splicing but also required for the export of mRNA out of the nucleus, even if the mRNA is encoded by an intron-less gene.

mRNA Export: RNP Remodeling by DEAD-Box Proteins
The DEAD-box protein Dbp5 was thought to remodel ribonucleoproteins and displace proteins from these complexes in an ATP-dependent fashion to allow for mRNA export from the nucleus. A recent study on Dbp5 shows, however, that its ADP-bound form may also perform an important function in displacing export factors from mRNA.

Transduction of developmental and environmental cues into the nucleus to induce transcription and the export of RNAs to the cytoplasm through the nuclear pore complex (NPC) play pivotal roles in regulation of gene expression. The process of bulk export of mRNAs from nucleus to cytoplasm is highly conserved across eukaryotes. Assembly of export-competent mRNA ribonucleoprotein (mRNP) is coupled ...
متن کاملA role for Gle1, a regulator of DEAD-box RNA helicases, at centrosomes and basal bodies
Control of organellar assembly and function is critical to eukaryotic homeostasis and survival. Gle1 is a highly conserved regulator of RNA-dependent DEAD-box ATPase proteins, with critical roles in both mRNA export and translation. In addition to its well-defined interaction with nuclear pore complexes, here we find that Gle1 is enriched at the centrosome and basal body. Gle1 assembles into th...
متن کاملDbp5, a DEAD-box protein required for mRNA export, is recruited to the cytoplasmic fibrils of nuclear pore complex via a conserved interaction with CAN/Nup159p.
Dbp5 is a DEAD-box protein essential for mRNA export from the nucleus in yeast. Here we report the isolation of a cDNA encoding human Dbp5 (hDbp5) which is 46% identical to yDbp5p. Like its yeast homologue, hDbp5 is localized within the cytoplasm and at the nuclear rim. By immunoelectron microscopy, the nuclear envelope-bound fraction of Dbp5 has been localized to the cytoplasmic fibrils of the...
متن کاملPi Release Limits the Intrinsic and RNA-Stimulated ATPase Cycles of DEAD-Box Protein 5 (Dbp5)
mRNA export from the nucleus depends on the ATPase activity of the DEAD-box protein Dbp5/DDX19. Although Dbp5 has measurable ATPase activity alone, several regulatory factors (e.g., RNA, nucleoporin proteins, and the endogenous small molecule InsP6) modulate catalytic activity in vitro and in vivo to facilitate mRNA export. An analysis of the intrinsic and regulator-activated Dbp5 ATPase cycle ...
